Sphingosine-1-phosphate (S1P) is a bioactive sphingolipid metabolite that is generated by the phosphorylation of sphingosine by sphingosine kinase 1 or 2. It is present in low concentrations in tissues such as lymphoid organs and thymus but is present in high concentrations in the blood, usually bound to ApoM in HDL particles. It acts as an intracellular second messenger as well as an extracellular ligand for G-protein-coupled receptors. S1P is implicated in a large number of physiological and cellular processes such as cell survival, immune cell trafficking, apoptosis, atherosclerosis, inflammation, angiogenesis, and tumorigenesis. Thus S1P could serve as a promising target for the treatment of human diseases. 96Trace’s Sphingosine-1-phosphate (Human) ELISA kit is based on the Competitive ELISA principle to quantitatively measure the concentration of Sphingosine-1-phosphate in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. The HRP-Streptavidin is added and any unattached conjugates are washed off using Wash Buffer. The HRP enzymatic reaction is detected by the addition of TMB-substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is inversely proportional to the concentration of S1P in the sample or standard.
