Src homology region 2 (SH2) domain-containing protein tyrosine phosphatase-2 (SHP2) is a non-receptor phosphotyrosine phosphatase. Ubiquitous expression of SHP2 is observed in a variety of cells and tissues. It is primarily localized in the cytoplasm but may also be present in other organelles (nucleus, mitochondria). SHP-2 is involved in multiple signal transduction pathways activated by hormones, cytokines, growth factors, and extracellular matrix proteins. Additionally, SHP2 plays a significant role in cell survival, differentiation, growth, and migration. When in the nucleus, SHP2 promotes DNA damage-induced apoptosis. SHP-2 mutations are associated with developmental disorders such as Noonan syndrome and LEOPARD syndrome. SHP2 is also involved in the pathogenesis of other diseases such as neurodegenerative, skeletal, metabolic diseases, and cancer. Thus, SHP-2 could serve as an efficient therapeutic target for the treatment of human diseases. 96Trace’s SHP2 (Human) ELISA kit is based on the Sandwich ELISA principle to quantitatively measure the concentration of SHP2 in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. The HRP-Streptavidin is added and any unattached conjugates are washed off using Wash Buffer. The HRP enzymatic reaction is detected by the addition of TMB-substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is directly proportional to the concentration of SHP2 in the sample or standard
