Receptor interacting protein kinase 3 (RIPK3) is a protein that belongs to the receptor-interacting protein (RIP) family of serine/threonine protein kinases. This cytosolic protein contains an active kinase domain at the N-terminus and a unique homotypic protein-protein interaction domain called RIP homotypic interaction motif (RHIM) at the C-terminus. RIPK3 interacts with RIPK1 via the RHIM domain resulting in the activation of RIPK3 and subsequent phosphorylation of MLKL. Phosphorylation results in a conformational change and promotes MLKL oligomerization. MLKL translocates to the plasma membrane and causes membrane disruption, ultimately resulting in necroptotic cell death. RIPK3 is also known to promote host defense against multiple DNA and RNA viruses such as Herpes simplex viruses (HSV), cytomegaloviruses (CMV), and Influenza A viruses (IAV). RIPK3 also plays a significant role in inflammation. Reduced expression of RIPK3 has been reported in several cancers such as breast, colorectal, cancer, melanoma, and acute myeloid leukemia (AML). Thus, RIPK3 may serve as an efficient target for anti-viral, anti-cancer, and anti-inflammation therapies. 96Trace’s RIPK3 (Human) ELISA kit quantitatively measures RIPK3 in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of RIPK3 in the sample or standard.
