Protein-arginine deiminase type-4 (PADI4) is an enzyme that belongs to the family of peptidyl arginine deiminases. PADI4 is expressed in immune cells, uterus, bone marrow, and brain. Similar to PADI2, PADI4 also catalyzes the conversion of arginine residues of proteins to citrulline, in the presence of calcium ions. PADI4 also is known to catalyze the citrullination of histones and in turn, regulate transcription and organization of the chromatin. PADI4 mediated histone citrullination plays an important role in DNA damage. Upregulation of PADI4 levels are observed in multiple tumors such as hepatocellular carcinoma, uterine, ovarian colorectal and lung adenocarcinomas. 96Trace’s PADI4 (Human) ELISA kit is based on the Sandwich principle to quantitatively measure the concentration of PADI4 in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. The HRP-Streptavidin is added and any unattached conjugates are washed off using Wash Buffer. The HRP enzymatic reaction is detected by the addition of TMB-substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is directly proportional to the concentration of PADI4 in the sample or standard.
