Neopterin (NP) is a 253 Da pyrazinopyridine derivative that is produced from guanosine triphosphate by the action of the enzyme guanosine triphosphate cyclohydrolase I. Upon stimulation by interferon-γ, the pteridine derivative is produced by multiple cells such as activated monocytes, dendritic cells, and macrophages. It is also produced to a lesser extent by fibroblasts, vascular smooth muscle cells, and renal epithelial cells. Neopterin can be detected in a variety of samples such as serum, urine, CSF, saliva, and synovial fluid. Elevated levels are observed during allograft rejections, bacterial and viral infections, autoimmune diseases, and cancer. Thus, Neopterin could serve as a useful marker for the prognosis of human diseases. 96Trace’s Neopterin (Human) ELISA kit is based on the Competitive ELISA principle to quantitatively measure the concentration of Neopterin in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. The HRP-Streptavidin is added and any unattached conjugates are washed off using Wash Buffer. The HRP enzymatic reaction is detected by the addition of TMB-substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is inversely proportional to the concentration of Neopterin in the sample or standard.
