Mid-regional proadrenomedullin (MR-proADM) is a 48 amino acid fragment that cleaves from proADM in a 1:1 ratio with adrenomedullin (ADM). ADM belongs to the calcitonin peptide family and is widely expressed in tissues such as the heart, lung, kidney, blood vessels, bone, etc. It possesses immunomodulatory, bactericidal, diuretic, and vasodilatory functions. However, ADM is very unstable and has a short half-life, thereby making it difficult to measure plasma concentrations of ADM in an immunoassay. MR-proADM is more stable, has a half-life of several hours, and directly reflects the concentrations of ADM in blood. Hence, MR-proADM serves as a reliable and promising serological biomarker for diagnosis and prognosis of sepsis, neurological disorders, septic shock as well as organ failure. 96Trace’s MR-proADM (Human) ELISA kit is based on the Sandwich principle to quantitatively measure the concentration of MR-proADM in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. The HRP-Streptavidin is added and any unattached conjugates are washed off using Wash Buffer. The HRP enzymatic reaction is detected by the addition of TMB-substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is directly proportional to the concentration of MR-proADM in the sample or standard
