A sandwich ELISA kit for the quantitative measurement of Lumican in Plasma,Serum,Tissue lysate,Other biological fluids.
Lumican is a leucine-rich repeat proteoglycan that is present in cornea, but is only weakly expressed in cartilage and bone. It is most similar in sequence to fibromodulin, it also contains N-linked keratan sulfate, but lacks the tyrosine-rich sequence. In the cornea, Lumican not only interacts with collagen molecules to limit fibril growth, but also plays a critical role in the regular spacing of fibrils and acquisition of corneal transparency by virtue of its keratan sulfate-containing glycosaminoglycan side chains LDC. a 96-well plate was coated with capture antibody. And the biotin conjugated antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and washed with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding the acidic stop solution. The density of color is proportional to the amount of Lumican captured from the samples.
