Heat shock 70-kDa protein 5 (HSPA5, GRP78, BIP) is a molecular chaperone protein that belongs to the heat shock protein 70 kDa family. HSPA5 acts as a master regulator of ER homeostasis. It is localized in the lumen of the endoplasmic reticulum (ER) and ensures proper folding and assembly of proteins in the ER. It binds to misfolded proteins to prevent aggregation of proteins, helps to translocate secretory proteins, and initiates unfolded protein response (UPR). Increased expression of HSPA5 is associated with viral infections. HSPA5 either contributes to the maturation of viral envelope proteins or directly participates in the entry of viruses into the host cells. Dysregulation of HSPA5 is associated with several diseases such as rheumatoid arthritis, atherosclerosis, Alzheimer's disease, and Type 2 diabetes. Therefore, HSPA5 may be a promising target for the treatment of human diseases. 96Trace’s HSPA5 (Human) ELISA kit quantitatively measures HSPA5 in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of HSPA5 in the sample or standard.
