A Sandwich ELISA kit for the Quantitative measurement of free Daratumumab in human serum or plasma samples

Daratumumab is a human IgG1 monoclonal antibody that binds with a high affinity to a unique epitope of CD38. CD38 is a type II transmembrane glycoprotein expressed on lymphoid, non-hematopoietic tissues, and myeloid cells. CD38 possesses ectoenzyme activity and plays a significant role in receptor-mediated regulation of cell adhesion and signal transduction. It is expressed at high levels on multiple myeloma cells, thus making it a unique target for therapeutic antibodies targeting multiple myeloma cell surfaces. The monoclonal antibody binds to CD38 on the surface of multiple myeloma cells and induces cell death via multiple mechanisms: complement-dependent cytotoxicity (CDC), antibody-dependent cell-mediated cytotoxicity (ADCC), antibody-dependent cellular phagocytosis (ADCP), and apoptosis. The original antibody is approved to treat multiple myeloma in patients who have received at least 3 previous therapies. Daratumumab ELISA kit measures free Daratumumab concentration in human serum or plasma with high sensitivity and reproducibility. The kit is based on the Sandwich ELISA principle. Standards and samples (serum or plasma) are added to the microtiter plate coated with the reactant for Daratumumab. After incubation, the wells are washed. The HRP conjugated probe is added and binds to Daratumumab captured by the reactant on the surface of the wells. Following incubation, wells are washed and the enzymatic activity is detected by the addition of TMB chromogen substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is proportional to the amount of Daratumumab in the sample or standard. The sample results can be determined directly using the standard curve.
