A Sandwich ELISA kit to quantitatively measure CNR1 in human serum, plasma, tissue lysates, and other biological fluids
Cannabinoid receptor 1 (CNR1, CB1) is a G-protein coupled cannabinoid receptor and an important component of the endocannabinoid system. CNR1 is primarily expressed in the central and peripheral nervous system, especially on the presynaptic terminals and axon terminals in the cerebellum, frontal cortex, amygdala, hippocampus, hypothalamus, and midbrain. CNR1 is also expressed on cells of the pituitary gland, thyroid gland; fat, muscle and liver cells, sperm cells, and the retina. One of the main functions of CNR1 is to inhibit the release of inhibitory and excitatory neurotransmitters such as GABA, acetylcholine, norepinephrine, and dopamine. Another function includes binding of activated CNR1 to Gi/o-protein and thereby inhibit adenylate cyclase and reduce intracellular cAMP levels. The use of CNR1 agonists has been demonstrated to relieve pain (of neuropathic origin) and anxiety. Although the exact mechanism is unknown, CNR1 is known to play a role in cancer. 96Trace’s CNR1 (Human) ELISA kit is based on the Sandwich ELISA principle to quantitatively measure the concentration of CNR1 in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. The HRP-Streptavidin is added and any unattached conjugates are washed off using Wash Buffer. The HRP enzymatic reaction is detected by the addition of TMB-substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is proportional to the concentration of CNR1 in the sample or standard.
