A Sandwich ELISA kit to quantitatively measure Asprosin in human serum, plasma, tissue lysates, and other biological fluids

Asprosin is a novel glucogenic adipokine that plays a critical role in glucose homeostasis. The protein is formed as a result of Furin/PACE (paired basic amino acid cleaving enzyme) serine endopeptidase mediated cleavage of the C-terminal domain of profibrillin. It is predominantly produced and secreted by white adipose tissue. Plasma Asprosin levels oscillate in a circadian manner; the levels are low during feeding and high during fasting. One of the main functions of Asprosin is to stimulate the production and release of glucose in the liver via the CREB pathway. Additionally, in the brain, the protein stimulates the activity of AgRP neurons and promotes increased appetite. Elevated Asprosin expression occurs in disorders such as polycystic ovary syndrome (POCS), type 2 diabetes, and obesity. Antibody-based treatment of Asprosin is known to reduce glucose and insulin levels in diabetic mice. Thus, Asprosin could serve as an efficient therapeutic target to treat metabolic disorders. 96Trace’s Asprosin (Human) ELISA kit is based on the Sandwich ELISA principle to quantitatively measure the concentration of Asprosin in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. The HRP-Streptavidin is added and any unattached conjugates are washed off using Wash Buffer. The HRP enzymatic reaction is detected by the addition of TMB-substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is proportional to the concentration of Asprosin in the sample or standard.
