Triggering receptor expressed on myeloid cells 2 (TREM2) is a single-pass cell surface transmembrane glycoprotein that belongs to the superfamily of immunoglobulins. It is expressed in macrophages, dendritic cells, myeloid cells, and osteoclasts. TREM2 binds to ligands such as bacterial LPS and DNA, phospholipids, and sulfatides. TREM2 also binds to transmembrane adaptor proteins such as DNAX-activation protein 12 (DAP12) and induces phagocytosis of pathogens and cell debris. TREM2 acts as an anti-inflammatory molecule and decreases the expression and secretion of pro-inflammatory cytokines such as TNFα and IL-6. TREM2 also regulates the cell number, proliferation, and survival of myeloid cells. Mutations in TREM2 result in diseases such as dementia, Alzheimer’s disease, ALS, Parkinson’s disease, and Nasu-Hakola disease. 96Trace’s TREM2 (Human) ELISA kit is based on the Sandwich principle to quantitatively measure the concentration of TREM2 in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. The HRP-Streptavidin is added and any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected by the addition of TMB-substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is directly proportional to the concentration of TREM2 in the sample or standard.
