Glucagon-like peptide-1 receptor (GLP-1R) is a 463 amino acid receptor protein that belongs to the class B family of G protein-coupled receptors (GPCRs). GLP1R is in abundance on the β-cells of the pancreas and neurons. However, it is also present in the heart, lung, kidney, blood vessels, and lymphocytes. In the brain, GLP1R regulates appetite; it also protects the neuronal cells from oxidative injury, and overexpression in mice brain promotes improved learning abilities. On the other hand, GLP1R drives glucose-dependent insulin secretion in the β-cells of the pancreas. Activated GLP1R stimulates the adenylyl cyclase pathway, which in turn promotes the synthesis and release of insulin. Due to its critical role in insulin secretion, production of hepatic glucose, food intake, and body weight, GLP1R is considered a promising target for the therapeutic treatment of obesity and diabetes mellitus. 96Trace’s GLP1R (Human) ELISA kit quantitatively measures GLP1R in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of GLP1R in the sample or standard.
