Aryl hydrocarbon receptor nuclear translocator-like protein 1 (ARNTL, BMAL1) is a basic helix-loop-helix transcription factor with 2 PAS domains. It is a critical component of the circadian clock. ARNTL contributes to the positive limb of the transcription-translation feedback loop. It heterodimerizes with CLOCK and induces the expression of downstream circadian clock genes Periods (Pers) and Cryptochromes (Crys) inside the nucleus. Upon synthesis in the cytoplasm, PER and CRY proteins bind to Casein Kinases (CKs), translocate back to the nucleus, and inhibit CLOCK-ARNTL transcriptional activity. ARNTL also regulates hypertension, diabetes, and obesity. Mutations and knockouts of ARNTL are associated with infertility, impaired glucose and lipid homeostasis, disturbed sleep patterns, reduced life span, and age-related pathologies. 96Trace’s ARNTL (Human) ELISA kit quantitatively measures ARNTL in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of ARNTL in the sample or standard.
