Sodium-dependent glucose cotransporter 2 (SGLT2) is an integral membrane protein that belongs to the SGLT family of glucose transporters. It is expressed in the early proximal convoluted tubule in the kidneys and is responsible for the reabsorption of 90% of glucose from the glomerular filtrate. SGLT2 mutations are associated with a rare autosomal recessive disorder called familial renal glucosuria (FRG). Patients suffering from this disorder excrete excess amounts of glucose in the urine despite having normal blood glucose levels. Due to the critical role of SGLT2 in glucose reabsorption, current treatment strategies involve the use of inhibitors to SGLT2 for the treatment of Type 2 diabetes. 96Trace’s SGLT2 (Human) ELISA kit quantitatively measures SGLT2 in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected by the addition of TMB-substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is directly proportional to the amount of SGLT2 in the sample or standard.
