Arachidonate 15-lipoxygenase (ALOX15) is a non-heme iron-containing dioxygenase enzyme that plays a critical role in the metabolism of polyunsaturated fatty acids. ALOX15 oxidizes omega-6 and omega-3 fatty acids to generate bioactive lipid metabolites such as eicosanoids, lipoxins, resolvins, and protectins. It is highly expressed in eosinophils, reticulocytes, alveolar macrophages, and dendritic cells. ALOX15 and its metabolites are implicated in the pathophysiology of multiple inflammatory diseases such as sepsis, arthritis, asthma, cystic fibrosis, and atherosclerosis. ALOX15 plays a dual role in carcinogenesis: it acts as a tumor suppressor in the lung, breast, head and neck, and esophageal cancer, whereas it is oncogenic in prostate and colorectal cancer. 96Trace’s ALOX15 (Human) ELISA kit quantitatively measures ALOX15 in human serum, plasma, and other biological fluids. The kit is based on the Sandwich ELISA principle. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. The HRP-Streptavidin is added and any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected by the addition of TMB-substrate. Finally, the reaction is terminated with an acidic stop solution. The color developed is directly proportional to the amount of ALOX15 in the sample or standard.
