Hypoxia-inducible factor 1-alpha (HIF1 alpha) functions as a master transcriptional regulator of the adaptive response to hypoxia. Under hypoxic conditions, stabilized HIF1 alpha translocate to the nucleus and promotes the transcription of a host of genes that enable the cell to adapt to the lack of oxygen. HIF1 alpha plays an essential role in embryonic vascularization, tumor angiogenesis and pathophysiology of ischemic disease. 96Trace’s HIF1 alpha (Mouse) ELISA kit is based on Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to mouse HIF1 alpha. Standards or samples are added to the micro ELISA plate wells that bind to the specific antibody. Then a biotinylated detection antibody specific for mouse HIF1 alpha and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. The wells are washed, a TMB substrate solution is added to the wells and blue color develops in proportion to the amount of HIF1 alpha bound. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of mouse HIF1 alpha. The concentration of mouse HIF1 alpha in the samples can be calculated by comparing the OD of the samples to the standard curve.
