Cryptochrome 1 (CRY1) is a photolyase-like-blue light receptor that belongs to the class of flavoproteins. It was first identified in Arabidopsis and later discovered in other lineages. In Arabidopsis, CRY1 mediates inhibition of hypocotyl elongation in response to blue light and photo periodically control floral initiation. In Drosophila, CRY1 binds to TIM in a light-dependent manner and mediates its degradation. In mammals, CRY1 heterodimerizes with PERIOD proteins, enters the nucleus, and suppresses CLOCK-BMAL1 transcriptional activity in a light-independent manner, thereby forming a negative feedback loop of the mammalian circadian clock. Additional functions of CRY1 include the regulation of glucose homeostasis and proinflammatory cytokine production. CRY1 is also known to downregulate the cAMP/PKA signaling pathway and promote cell growth and proliferation. Overexpression of CRY1 is associated with chronic lymphocytic leukemia and colorectal cancers and thus may serve as a promising target for anticancer therapy. 96Trace’s CRY1 (Human) ELISA kit quantitatively measures CRY1 in human serum, plasma, and other biological fluids. Test samples, Standards, and Biotinylated Detection antibody are added to the wells pre-coated with capture antibody and then washed with Wash Buffer. HRP-Streptavidin is added to the plate, and post-incubation any unattached conjugates are washed off by Wash Buffer. The HRP enzymatic reaction is detected using TMB-substrate. Finally, an acidic stop solution terminates the enzymatic reaction. The color developed is directly proportional to the amount of CRY1 in the sample or standard.
